Retatrutide for Weight Loss
GLP-1 / GIP / Glucagon injection
Retatrutide is a next-generation weight loss medication that activates three key metabolic pathways: glucagon-like peptide-1 (GLP-1), glucose-dependent insulinotropic polypeptide (GIP), and glucagon receptors. This triple-agonist approach helps regulate hunger, energy use, and blood sugar more powerfully than previous medications. Retatrutide also:

about Retatrutide

What is Retatrutide?

Retatrutide is an investigational drug that targets three key hormone pathways—GLP-1, GIP, and glucagon receptors—to support both blood sugar control and weight loss. It is currently being studied as a next-generation treatment for obesity and type 2 diabetes.

While not yet FDA-approved, clinical trials have shown promising results for significant weight reduction, improved insulin sensitivity, and enhanced metabolic function. Retatrutide may offer greater benefits than single or dual hormone agonists by combining multiple pathways in one therapy.

Retatrutide is administered as an injection and is still under evaluation for safety and efficacy. As of now, there is no FDA-approved or generic version of retatrutide available.

Benefits of Retatrutide

Expected weight loss with Retatrutide is dose-dependent. In early clinical trials, participants using higher doses of Retatrutide experienced significant weight reductions, with some studies reporting over 24% total body weight loss, potentially exceeding results from current GLP-1 therapies.

Retatrutide may also support metabolic and cardiovascular health by improving markers such as blood pressure, cholesterol, insulin sensitivity, and blood sugar levels. Its triple-agonist action (GLP-1, GIP, and glucagon) is designed to enhance both fat burning and appetite control more effectively than dual-agonist drugs.

Additionally, many patients have reported reduced cravings, a quieter mind around food (“food noise”), and better energy, mood, and overall quality of life during the course of therapy.

Benefits of Retatrutide

When you eat, your body releases gut hormones like GLP-1, GIP, and glucagon. These help regulate blood sugar, slow digestion, signal fullness, and manage how the body stores or burns fat. Retatrutide activates all three of these hormone pathways, offering a broader and potentially more powerful metabolic response than GLP-1 medications alone.

In some people, these natural hormones aren’t released efficiently—or the brain doesn’t respond to them as it should.

Retatrutide is a triple receptor agonist (RA) that mimics GLP-1, GIP, and glucagon. It interacts with the same receptors your body uses, helping regulate appetite, slow digestion, and increase energy expenditure. This combination promotes weight loss, improves insulin sensitivity, and supports long-term metabolic health.

Because Retatrutide supports biological processes that may be underperforming, consistent use over time may be necessary for optimal results.

It’s important to understand that Retatrutide won’t work the same for everyone. There are many reasons why weight loss can be difficult. At Out Of Pounds, our clinicians specialize in uncovering the root causes of weight gain and personalizing treatment based on your needs. Every prescription is determined by a licensed provider following an in-depth evaluation.

More on personalized treatment with OOP

How to use it

Retatrutide is typically administered as a once-weekly injection. Always follow your healthcare provider’s instructions carefully when using this medication. In clinical trials, retatrutide has been delivered via subcutaneous injection—commonly in the upper arm, thigh, or abdominal area.

Since retatrutide is still in development, proper dosing and storage instructions will be provided once approved. If using a prefilled pen or vial, follow handling guidelines and keep it refrigerated unless otherwise directed.

Dosing

Dosing will depend on your personalized prescription, medical history, and provider’s evaluation. Always follow medical advice and packaging guidelines closely.

Side Effects

Common side effects may include nausea, diarrhea, decreased appetite, or constipation. These symptoms often improve over time. Less common effects can include dizziness, fatigue, or stomach discomfort. Report any unusual symptoms to your provider immediately.

Black Box Warning

As of now, Retatrutide does not carry an FDA-approved black box warning, but given its similarity to other GLP-1 agonists, potential thyroid C-cell tumor risks may be evaluated during approval. Discuss risks and benefits with your provider.

The FDA also warns not to take Zepbound or Mounjaro if you have a personal or family history of multiple endocrine neoplasia syndrome Type 2 or medullary thyroid cancer.

Precautions

Retatrutide should be used with caution in individuals with a history of pancreatitis, thyroid disorders, or gastrointestinal disease. It is not recommended for use during pregnancy or while breastfeeding unless medically necessary.

Interactions

Tell your provider about all medications you’re taking. Retatrutide may delay gastric emptying, which could impact how your body absorbs oral medications, especially those that require fast absorption.
